Is it possible to create custom layouts on Production Studio?

You can use OBS Studio and its window capture feature to reformat Zoom when streaming to YouTube or Facebook. There are also templates available in share screen's layouts and options. There are also templates under immersive view. If you want to have the 2 tiles the same size you will lose significant resolution of any displayed text (each tile is now 640 x 360), but if it still works you could use 2 separate devices. One devices to display the presentation via OBS Studio' s virtual camera and window capture feature (not using share screen). The second device is the speaker. The host then sets both videos as spotlighted. Keep in mind that this is not a best practice for presentation quality.

While @storyhub provided a workaround for you, he didn’t specifically answer your question. 

The answer, of course, is No. Production Studio scenes can only be created within the existing scene framework, and only edited within the Events or Web Portal. Production Studio isn’t designed to be infinitely configurable, and to keep the functionality simple enough that non-technical people can still operate it, there’s definitely some trade offs that have to be made.

If you need more flexibility, solutions like OBS Studio, vMix (Windows), eCamm or Mimo Live (MacOS) can be used with a lot of flexibility — as long as you’re willing to drive the learning curve.
